If you are talking about the in-game "100%", visit our 100% checklist. The in-game "100%" serves only as an achievement activator and covers only a portion of everything possible in Silksong.
You can get everything for 100% in the first run, but you should aim to do so on Steel Soul Mode to get all possible unlocks. Steel Soul Mode is only accessible after beating the game once.
The absolute minimum to get 100% Completion and All Achievements in Hollow Knight: Silksong is therefore 2 optimized runs. Read our guide: How to get all achievements.
The catch is that for several achievements, you need to get 100% Completion while on Steel Soul mode and under 30 hours, while the first section of the game needs to be completed under 5 hours.
Steel Soul Mode is only accessible after beating the game the first time, so have a normal, non-spoiler run, then another very optimized and skilled run for all Steel Soul, Speedrun and Completion achievements. This is hard and will likely require more attempts.
60 hours in one world. If you aim for all achievements and Steel Mode, that will take another 20 hours for a successful run and likely many hours in lost Steel Soul runs.
10/10, considering all the achievements and the nature of Steel Soul. It is possible and not as hard as it sounds, but still very punishing.
There are also other really complicated items like Witch Crest or the last Pale Oil.
Silksong is in its nature all free-roam. Go wherever you want, anytime you want.
The most convenient time to do it would be after acquiring all possible movement upgrades and before entering Act 3. But a few hours of collecting are still necessary in Act 3.

What kind of data does the progress tracking functionality collect?
This page only displays your progress. The checklists you use send database keys for checked items plus a timestamp. That is tied to your user ID and account email. Sign-in and storage use a cloud database.
Does this support DLC?
Silksong: Sea of Sorrow will be supported at launch. Expedition 33 DLC is included in our checklists. The Witcher 3 and Kingdom Come: Deliverance 2: full DLC coverage is planned very soon. Hades 2 and Nine Sols: no DLC exists yet.
